  • PTHR ID & PTN node:
    PTN008143312

  • Sequences with problematic annotation
    UniProt Q9UBN7 (human HDAC6)

  • Type of Issue: Erroneous source or erroneous propagation, or other issue
HDAC6 was originally thought to be a histone deacetylase based on papers such as PMID:10220385, an early cloning paper which showed histone deacetylation activity in vitro. Since then, it has been shown that HDAC6 does not acetylate histones in vivo with many papers showing that it is a predominantly cytoplasmic protein which deacetylates various non-histone substrates.

  • Add the label 'high priority' if needed. Generally high priority issues affect a lot of proteins, and annotations are incorrect (as opposed to just imprecise).

Hello @magrane
Thanks a lot for this information. It is very interesting since the HDAC6 subfamily clusters with the HDAC10 subfamily for which histone deacetylase activity was also shown in vitro but refuted subsequently. Both are cytoplasmic. So I've reviewed the annotation for the whole node PTN008492157. Did you update the primary GO annotation ?
Cheers,
Marc

Hi Marc,
Thanks for looking into this. I've updated the UniProt HDAC6 records (changes are already available internally) and have fixed the primary GO annotations for these.

This is perfect. It is a nice improvement of the annotation of this complex family.
Thanks again.
Marc.
